미토콘드리아 DNA 중합효소는 보조 소단위의 동형 이합체와 촉매 소단위로 구성된 이종삼량체입니다. 이 유전자에 의해 인코딩되는 단백질은 미토콘드리아 DNA 중합효소의 촉매 소단위입니다. 인코딩된 단백질은 N-말단 근처에 다형성일 수 있는 폴리글루타민 트랙트를 포함합니다. 이 유전자의 결함은 미토콘드리아 DNA 결실 1을 동반하는 진행성 외안근마비(PEOA1), 감각성 운동실조성 신경병증 구음장애 및 안근마비(SANDO), 알퍼스-후텐로허 증후군(AHS), 그리고 미토콘드리아 신경위장관 뇌병증 증후군(MNGIE)의 원인이 됩니다. 이 유전자에 대해 동일한 단백질을 인코딩하는 두 가지 전사 변이체가 발견되었습니다.
