이 유전자는 발달 중인 뼈, 사지, 털, 치아 및 유방 조직의 중배엽에서 발현되는 쌍-유사 호메오도메인 전사 인자를 인코딩합니다. 이 유전자의 돌연변이는 두정골의 불완전한 골화를 특징으로 하는 상염색체 우성 질환인 두정골 구멍 2 (PFM2)를 유발합니다. 이 유전자의 돌연변이는 또한 탈모증 및 성선기능저하증을 동반하는 전두비강 이형성증의 한 형태를 유발하며, 이는 두개안면 발달, 중배엽-상피 세포 간 통신 및 모낭 발달에서 이 유전자의 역할을 시사합니다. 이 유전자를 포함하는 염색체 11번의 일부 결실, del(11)(p11p12)은 포토키-샤퍼 증후군 (PSS)을 유발합니다. 이 증후군은 두개안면 이상, 인지 장애, 다발성 외골종 및 남성의 생식기 이상을 특징으로 합니다. 마우스에서는 이 유전자가 16개 코돈 떨어진 두 개의 이중 번역 개시 부위를 사용하는 것으로 나타났습니다.
