이 유전자는 다효소 피루브산 탈수소효소 복합체(PDC)의 E2 구성 요소를 인코딩합니다. PDC는 미토콘드리아 내막에 존재하며 피루브산을 아세틸 조효소 A로 전환하는 반응을 촉매합니다. 이 유전자의 단백질 산물인 다이하이드로리포아마이드 아세틸트랜스퍼라제는 피루브산의 산화적 탈카르복실화에 의해 형성된 아세틸기를 받아 조효소 A로 전달합니다. 다이하이드로리포아마이드 아세틸트랜스퍼라제는 항미토콘드리아 항체의 항원입니다. 이러한 자가항체는 자가면역 간 질환인 원발성 담즙성 간경변증(PBC) 환자의 거의 95%에서 발견됩니다. PBC에서는 활성화된 T 림프구가 이 단백질이 비정상적으로 분포하고 과발현되는 담관의 상피 세포를 공격하고 파괴합니다. PBC는 결국 간경변증과 간부전으로 이어집니다. 이 유전자의 돌연변이는 또한 영아 및 유아기에 원발성 젖산증을 유발하는 피루브산 탈수소효소 E2 결핍증의 원인이 됩니다.
