TGFB3는 TGF-베타(transforming growth factor-beta) 초패밀리에 속하는 분비성 리간드(ligand)를 암호화하며, 이 리간드는 다양한 TGF-베타 수용체에 결합해 유전자 발현을 조절하는 SMAD 전사인자(transcription factor)의 동원과 활성화를 유도한다. 암호화된 전구단백질은 단백질분해 가공을 거쳐 잠재성 관련 펩타이드(latency-associated peptide, LAP)와 성숙 펩타이드를 형성하며, 배아 발생과 세포 분화에 관여하고 상처 치유에서도 역할을 할 수 있다. 이 유전자의 돌연변이는 대동맥류 및 대동맥 박리, 그리고 가족성 부정맥 유발성 우심실 이형성증(arrhythmogenic right ventricular dysplasia) 1형의 원인이 된다.
