유전자 개요

TGFB1은 TGF-베타(transforming growth factor-beta, TGF-β) 슈퍼패밀리(superfamily)에 속하는 분비성 리간드(ligand)를 암호화하며, 이 리간드는 여러 TGF-베타 수용체에 결합해 유전자 발현을 조절하는 SMAD 계열 전사인자(transcription factor)의 동원과 활성화를 유도한다. 암호화된 전구단백질은 단백질분해 처리를 거쳐 잠복 관련 펩타이드(latency-associated peptide, LAP)와 성숙 펩타이드로 가공되며, 성숙 펩타이드 동종이량체로만 이루어진 활성형 또는 LAP·잠복 TGF-베타 결합단백질과 결합한 잠복형으로 존재한다. 세포 증식·분화·성장을 조절하고 인터페론 감마(interferon gamma)와 종양괴사인자 알파(tumor necrosis factor alpha) 등 다른 성장인자의 발현과 활성도 조절하며, 종양 세포에서 흔히 상향 조절되고 이 유전자의 돌연변이는 카무라티-엥겔만병(Camurati-Engelmann disease)을 일으킨다.
