TFPI는 혈액 응고의 조직인자(tissue factor, TF) 의존 경로를 조절하는 Kunitz형 세린 단백질분해효소 억제제(serine protease inhibitor)를 암호화한다. 응고는 factor VIIa-TF 복합체 형성으로 시작되어 factor IX와 X를 단백질분해적으로 활성화하고 최종적으로 피브린(fibrin) 응괴 형성으로 이어지는데, TFPI 산물은 자가조절 루프에서 활성화된 factor X와 VIIa-TF를 억제한다. 이러한 억제는 혈우병(hemophilia) 동물 모델에서 지혈을 회복시키며, TFPI는 억제 활성·특이성·세포 내 위치가 서로 다른 여러 단백질 아이소폼(isoform)를 암호화한다.
