유전자 개요
이 유전자로 암호화되는 단백질은 초파리 유전자 ‘mothers against decapentaplegic’(Mad) 및 C. elegans 유전자 Sma의 유전자 산물과 유사한 단백질 계열인 SMAD 계열에 속한다. SMAD 단백질은 여러 신호 전달 경로를 매개하는 신호 전달자이자 전사 조절자이다. 이 단백질은 세포 성장, 세포자멸사(apoptosis), 형태 형성, 발달 및 면역 반응을 포함한 다양한 생물학적 활성에 관여하는 골형성단백질(BMPs)의 신호를 매개한다. BMP 리간드에 반응하여, 이 단백질은 BMP 수용체 키나아제(kinase)에 의해 인산화되고 활성화될 수 있다. 인산화된 형태의 이 단백질은 SMAD4와 복합체를 형성하며, 이는 전사 조절에서 기능하는 데 중요하다. 이 단백질은 SMURF1 및 SMURF2와 같은 SMAD 특이적 E3 유비퀴틴(ubiquitin) 리가아제의 표적이며, 유비퀴틴화(ubiquitination) 및 프로테아좀 매개 분해를 겪는다. 동일한 단백질을 암호화하는 대체 스플라이싱 전사체 변이체가 관찰되었다.
