RAB5는 GDP 결합 활성(GDP binding activity), GTP 결합 활성(GTP binding activity), 및 GTPase 활성(GTPase activity)을 가진다. 세포통과수송(transcytosis)에 의한 아밀로이드 베타(amyloid-beta) 제거, 초기 엔도솜(early endosome)에서 후기 엔도솜(late endosome)으로의 수송, 세포 외 유출(exocytosis) 조절 등 여러 과정에 관여한다. 초기 엔도솜 막의 세포질 측(cytoplasmic side of early endosome membrane), 핵질(nucleoplasm), 말단 부위(terminal bouton) 등 다양한 세포 구성요소에 위치한다.
