PVR가 암호화하는 단백질은 면역글로불린 슈퍼패밀리(immunoglobulin superfamily)에 속하는 막관통 당단백질(transmembrane glycoprotein)이다. 세포외 도메인은 세포외기질 분자인 비트로넥틴(vitronectin)에 대한 세포 부착을 매개하며, 세포내 도메인은 다이네인 경쇄(dynein light chain) Tctex-1/DYNLT1과 상호작용한다. 영장류 계통에 특이적으로 존재하고 폴리오바이러스(poliovirus) 복제의 첫 단계에서 세포 수용체로 작용하며, 서로 다른 아이소폼(isoform)을 암호화하는 여러 전사체 변이가 확인되었다.
