PKLR이 암호화하는 단백질은 포스포에놀피루브산(phosphoenolpyruvate)을 피루브산(pyruvate)과 ATP로 전환하는 인산기전이반응(transphosphorylation)을 촉매하는 피루브산 키나아제(pyruvate kinase)로, 이 반응은 해당과정(glycolysis)의 속도제한 단계이다. 유전자 돌연변이나 유전적 변이로 인한 이 효소의 결함은 만성 유전성 비구상적혈구 용혈성 빈혈(CNSHA 또는 HNSHA)의 흔한 원인이다. 서로 다른 아이소폼(isoform)을 암호화하는 여러 전사체 변이(transcript variant)도 보고되어 있다.
