PAPPA는 분비성 금속단백분해효소(metalloproteinase)를 암호화하며, 이 효소는 인슐린 유사 성장 인자 결합 단백질(insulin-like growth factor binding proteins, IGFBPs)을 절단한다. 절단 이후 인슐린 유사 성장 인자가 IGFBP에서 해리되어 IGF 수용체에 결합함으로써 IGF 경로가 활성화되며, 암호화된 단백질은 골 형성, 염증, 상처 치유 및 여성 생식능에 관여한다. 발현 증가는 당뇨병성 신증(diabetic nephropathy)과 연관되고, 여러 사람 암종에서 종양 침윤과 성장을 촉진할 수 있다.
