P53는 전사 활성화·DNA 결합·올리고머화(oligomerization) 도메인을 갖춘 종양 억제(tumor suppressor) 단백질을 암호화하며, 다양한 세포 스트레스에 반응해 표적 유전자의 발현을 조절한다. 이를 통해 세포 주기(cell cycle) 정지, 세포자멸사(apoptosis), 노화, DNA 복구, 대사 변화 등을 유도한다. P53 유전자의 돌연변이는 리-프라우메니 증후군(Li-Fraumeni syndrome) 같은 유전성 암을 포함한 다양한 인간 암과 연관되며, 대체 스플라이싱과 대체 프로모터·번역 개시 코돈 사용으로 여러 전사체 변이와 아이소폼(isoform)이 생성된다.
