이 유전자는 NOTCH 단백질 계열의 구성원을 암호화한다. 이 제1형 막관통 단백질 계열 구성원들은 세포외 도메인에 다수의 표피 성장 인자 유사(epidermal growth factor-like, EGF) 반복 서열과, 여러 다른 유형의 도메인으로 구성된 세포내 도메인을 포함하는 구조적 특징을 공유한다. Notch 신호 전달은 Notch 계열 수용체가 그에 상응하는 리간드에 결합함으로써 물리적으로 인접한 세포 간의 상호작용을 조절하는 진화적으로 보존된 세포 간 신호 전달 경로이다. 암호화된 전구 프로단백질은 트랜스-골지 망에서 단백질 분해를 거쳐 두 개의 폴리펩타이드 사슬을 생성하며, 이들은 이합체화되어 성숙한 세포 표면 수용체를 형성한다. 이 수용체는 혈관, 신장, 간 발달에 관여할 수 있다. 이 유전자의 돌연변이는 정신분열증과 관련될 수 있다. 대안적 스플라이싱은 다수의 전사체 변이를 초래하며, 이들 중 적어도 하나는 단백질 분해를 거치는 이소형을 암호화한다.
