NOTCH3는 초파리(Drosophila melanogaster)의 I형 막단백질 Notch에 대한 인간 상동체(homologue) 중 세 번째로 발견된 단백질을 암호화한다. 초파리에서 Notch는 세포 결합 리간드(ligand)인 delta 및 serrate와 상호작용하여 신경 발달에 핵심적 역할을 하는 세포 간 신호전달 경로를 형성하며, Notch 리간드의 인간 상동체도 확인되었으나 이들과 인간 Notch 상동체 사이의 정확한 상호작용은 아직 규명되지 않았다. NOTCH3의 돌연변이는 피질하 경색과 백질뇌병증을 동반하는 대뇌 상염색체 우성 동맥병증(CADASIL)의 근본 원인으로 밝혀졌다.
