MSTN은 TGF-베타(transforming growth factor-beta) 단백질 슈퍼패밀리(superfamily)에 속하는 분비성 리간드(ligand)를 암호화하며, 이 리간드는 여러 TGF-베타 수용체에 결합해 유전자 발현을 조절하는 SMAD 계열 전사인자(transcription factor)의 동원과 활성화를 유도한다. 암호화된 전구단백질(preproprotein)은 단백질 분해 가공을 거쳐 이황화 결합으로 연결된 동종이량체(homodimer)의 각 소단위를 형성하며, 생성된 단백질은 골격근 세포의 증식과 분화를 음성적으로 조절한다. MSTN의 돌연변이는 사람과 그 밖의 포유류에서 골격근량 증가와 연관되어 있다.
