HP는 전구전단백질(preproprotein)을 암호화하며, 이 단백질은 가공을 거쳐 알파 사슬(alpha chain)과 베타 사슬(beta chain)로 나뉜 뒤 사량체(tetramer)로 결합하여 합토글로빈(haptoglobin)을 형성한다. 합토글로빈은 유리 혈장 헤모글로빈(free plasma hemoglobin)에 결합해 분해 효소가 헤모글로빈에 접근하도록 하는 동시에, 신장을 통한 철 손실을 막고 헤모글로빈으로 인한 신장 손상을 방지한다. 해당 유전자나 그 조절 부위의 돌연변이는 무합토글로빈혈증(ahaptoglobinemia) 또는 저합토글로빈혈증(hypohaptoglobinemia)을 유발하며, 암호화된 단백질은 세균에 대한 항균 활성도 나타낸다.
