유전자 개요

HMGCR이 암호화하는 HMG-CoA 환원효소(HMG-CoA reductase)는 콜레스테롤 합성의 속도 제한 효소이며, 이 효소가 촉매하는 반응의 산물인 메발론산에서 유래한 스테롤 및 비스테롤 대사산물이 매개하는 음성 되먹임(negative feedback) 기전으로 조절된다. 포유류 세포에서는 일반적으로 LDL 수용체(LDL receptor)를 통한 저밀도 지질단백질(low density lipoprotein, LDL)의 내재화 및 분해로 생성된 콜레스테롤에 의해 억제된다. 환원효소의 경쟁적 억제제는 간에서 LDL 수용체 발현을 유도하여 혈장 LDL의 이화작용을 증가시키고, 죽상동맥경화증(atherosclerosis)의 중요한 결정 인자인 혈장 콜레스테롤 농도를 낮춘다.
