ESAM은 세포-세포 접착 매개 활성(cell-cell adhesion mediator activity)을 가능하게 하며, 양세포 치밀연접 조립(bicellular tight junction assembly), 세포-세포 접착, 액틴 필라멘트 중합 조절(regulation of actin filament polymerization) 등 여러 과정에 관여한다. 세포-세포 접합부(cell-cell junction)와 세포막(plasma membrane)에 위치하고, 단백질 함유 복합체(protein-containing complex)의 구성 요소로 작용한다.
