EPHA4는 단백질 티로신 인산화효소(protein-tyrosine kinase) 계열 중 에프린 수용체(ephrin receptor) 서브패밀리(subfamily)에 속하며, EPH 및 EPH 관련 수용체는 특히 신경계에서 발생 과정을 매개하는 데 관여하는 것으로 알려져 있다. 이 서브패밀리(subfamily)의 수용체는 일반적으로 단일 인산화효소 도메인과, 시스테인 풍부(Cys-rich) 도메인 및 2개의 피브로넥틴 III형 반복(fibronectin type III repeat)을 포함하는 세포외 영역을 가진다. 에프린 수용체는 세포외 도메인 서열의 유사성과 ephrin-A·ephrin-B 리간드에 대한 결합 친화도에 따라 두 그룹으로 나뉘며, EPHA4에서는 서로 다른 아이소폼(isoform)을 암호화하는 여러 전사체 변이가 확인되었다.
