ENO1은 포유류의 세 가지 에놀라아제(enolase) 동질효소 중 하나인 알파-에놀라아제를 암호화하며, 각 동질효소는 알파·감마·베타 소단위체 2개로 이루어진 동종이량체(homodimer)로서 해당과정(glycolysis) 효소로 기능한다. 알파-에놀라아제는 단량체 형태에서 수정체 구조 단백질인 타우-크리스탈린(tau-crystallin)으로도 작용하고, 대체 스플라이싱(alternative splicing)으로 생성되는 짧은 아이소폼(isoform)은 c-myc 프로모터(promoter)에 결합해 종양 억제자로 기능하는 것으로 보고되었다. 또한 알파-에놀라아제는 하시모토 뇌병증(Hashimoto encephalopathy)의 자가항원(autoantigen)으로도 확인되었다.
