ELN은 탄성섬유(elastic fiber)를 구성하는 두 성분 중 하나인 단백질을 암호화하며, 이 탄성섬유는 세포외기질(extracellular matrix)의 일부로서 심장, 피부, 폐, 인대, 혈관 등의 장기와 조직에 탄성을 부여한다. 암호화된 단백질은 글리신(glycine)과 프롤린(proline) 같은 소수성 아미노산이 풍부하여 라이신(lysine) 잔기 간 가교로 경계 지어진 가동성 소수성 영역을 형성하며, 그 분해산물인 엘라스토카인(elastokine)은 단핵구와 피부 섬유아세포의 이동·증식을 자극하고 암 진행에도 기여할 수 있다. 이 유전자의 결실과 변이는 대동맥판상부협착증(supravalvular aortic stenosis, SVAS) 및 상염색체 우성 피부이완증(cutis laxa)과 연관된다.
