CLDN18은 클라우딘(claudin) 계열 단백질을 암호화하며, 클라우딘은 긴밀연접 가닥(tight junction strands)을 구성하는 내재성 막단백질(integral membrane protein)이다. 긴밀연접 가닥은 상피 또는 내피 세포층 사이의 세포주위 공간으로 용질과 물이 자유롭게 통과하는 것을 막는 물리적 장벽으로 작용하며, 세포 극성과 신호전달(signal transduction) 유지에도 핵심적인 역할을 한다. CLDN18은 궤양성 대장염 환자에서 상향조절되고 침윤성 유관 선암종에서 높게 과발현되며, 위 세포에서는 PKC/MAPK/AP-1(단백질 키나아제 C/미토겐 활성화 단백질 키나아제/활성화 단백질-1) 의존성 경로가 그 발현을 조절하고, 서로 다른 아이소폼(isoform)을 암호화하는 선택적 스플라이싱 전사체 변이들이 확인되었다.
