CDH5는 카드헤린 상위계열(cadherin superfamily)에 속하는 고전형 카드헤린을 암호화하며, 합성된 전구단백질은 단백질 분해 가공을 거쳐 성숙한 당단백질(glycoprotein)로 전환된다. 이 칼슘 의존성 세포-세포 부착 분자는 5개의 세포외 카드헤린 반복 단위, 막관통 영역, 그리고 고도로 보존된 세포질 꼬리로 구성되며, 동종친화적(homophilic) 부착 능력을 부여함으로써 내피 부착연접(adherens junction)의 형성과 유지에 관여한다. 해당 유전자는 16번 염색체 장완(long arm)의 유전자 클러스터에 위치하며, 이 영역은 유방암과 전립선암에서 이형접합성 소실(loss of heterozygosity) 사건과 연관되어 있다.
