BARD1(BARD1)은 BRCA1의 N-말단(N-terminal) 영역과 상호작용하는 단백질을 암호화하며, BRCA1의 가장 잘 보존된 두 영역인 N-말단 RING 모티프(RING motif)와 C-말단 BRCT 도메인(BRCT domain)과 상동성을 공유하고 3개의 직렬 안키린 반복(ankyrin repeats)을 포함한다. BARD1과 BRCA1의 결합은 BRCA1의 종양유발성 아미노산 치환에 의해 교란되며, 이는 두 단백질 간 안정적 복합체 형성이 BRCA1의 종양 억제(tumor suppression)에 필수적일 수 있음을 시사한다. 이 단백질은 유방암 또는 난소암(ovarian cancer)에서 발암성 돌연변이의 표적일 수 있으며, 서로 다른 아이소폼(isoform)을 암호화하는 여러 선택적 스플라이싱 전사체 변이가 확인되었다.
