ADAMTS5 유전자는 ADMTS(a disintegrin and metalloproteinase with thrombospondin motifs) 단백질 계열 구성원을 암호화한다. 이 계열 구성원들은 프로펩타이드 영역, 메탈로프로테이나제 도메인, 디스인테그린 유사 도메인, 그리고 트롬보스폰딘 제1형(TS) 모티프를 포함한 몇 가지 고유한 단백질 모듈들을 공유한다. 이 효소는 두 개의 C-말단 TS 모티프를 포함하며, 연골의 주요 프로테오글리칸인 애그리칸(aggrecan)을 절단하는 애그리칸아제(aggrecanase)로 작용한다. 이는 골관절염(osteoarthritis)에서 연골 파괴를 매개할 수 있다.
